Yes, Content Manager for iSeries Version 5 Release 3 (5.3) is supported for use on both i5/OS V5R4 (5722-SS1) and i5/OS V6R1 (5761-SS1).

This includes support for the ImagePlus® Workfolder Application Facility (WAF).
